IDENTIFICATION AND CHARACTERIZATION OF GENETIC FACTORS CONTRIBUTING TO CHRONIC KIDNEY DISEASE
Project goals
The aim of the project is to extend our currently limited knowledge on genetic and molecular basis of chronic kidney diseases (CKD). The project is based ? in agreement with currently accepted common disease-rare variants hypothesis ? on systematic identification of rare genetic variants in families and simplex cases with evidently genetically based kidney disease and their subsequent detailed molecular biology characterization. The research should define novel causal genes and basic pathogenetic effects of identified genetic variants on studied phenotypes and kidney biology in general. Better knowledge on genetic component and molecular basis of studied kidney diseases should allow proper diagnosis, prevention, targeted treatment and decrease in morbidity and mortality of CKD.

Identifikace a charakterizace genetických faktorů chronického onemocnění ledvin

Cílem projektu je rozšíření základních znalostí o genetické a molekulárně biologické podstatě chronického onemocnění ledvin. Projekt je - v souladu s hypotézou kauzality kombinace relativně vzácných funkčně významných variant v omezeném počtu genů - založen na systematickém vyhledávání a vzácných genetických variant v rodinách a sporadických případech s evidentně geneticky podmíněným chronickým onemocněním ledvin, následné molekulárně biologické charakterizaci nalezených genetických variant a definici základních biologicko-patologických procesů spojených se studovanými fenotypy. Hlubší znalost genetické komponenty CKD, možnosti jejího laboratorního určení a správná klinicko-biologická interpretace by měly významně přispět ke správné a včasné diagnose,prevenci, cílené terapii a snížení morbidity a mortality spojené s CKD.

The aim of the project was to extend our currently limited knowledge on genetic and molecular basis of chronic kidney diseases (CKD). Within the project we identified genes and mutations causing MCKD1 and several other not yet characterised familiar nephropathies.
