Dynamika aktinového cytoskeletu ve vztahu ke kvasinkové NADPH oxidáze YNO1, redox potenciálu a buněčnému cyklu.
Annotation in Czech
Cílem projektu je objasnit úlohu oxidativních radikálů v regulaci aktinového cytoskeletu v buněčné odpovědi na stres. V projektu je navrhováno využití kombinace metod fluorescenční mikroskopie živých buněk a imunoelektronové mikroskopie buněk fixovanýchza vysokého tlaku k analýze změn distribuce aktinu v kvasince S.cerevisiae. V rámci projektu budou vytvořeny specifické kmeny divokého typu či delečních mutantů (např. yno1 a yhm2) produkující známé aktinové markery (Abp1 a Abp140) fúzované s fluorescenčními proteiny (GFP, mRFP nebo mCherry). Pomocí těchto kmenů bude analyzován vliv teplotního, oxidativního a nutričního stresu na aktinový cytoskelet.
