Bioinspired Nanocomposite Structures for bone Tissue Regeneration
Project goals
Bioinspired composite materials promoting the regeneration of defective bone tissue will be prepared. Their proposed composition will imitate the real bone structure, and will combine the advantages of nanofibres, macroporous gels and bioactive inorganiccomponent. This materials will be composed of (1) organic matrix represented by relatively quickly degradable synthetic macroporous hydrogels based on methacrylates, macroporous hydrogel prepared from the natural collagen (both further supplemented with hyaluronic acid), (2) organic fibrous component represented by collagen or polylactide nanofibre scaffolds, and (3) inorganic component ? natural or synthetic nanocrystalline hydroxyapatite or tricalcium phosphate. All composite types will be mechanically tested. The effects of the composite materials as well as their individual components on the adhesion, growth, maturation, viability and potential immune activation of osteogenic cells (in vitro tests) will be studied.

Cílem tohoto grantového projektu je příprava biologicky insipirovaného nanokompozitního materiálu pro obnovu kostní tkáně, který bude imitovat reálnou strukturu kosti a zároveň kombinovat výhody nanovláken, makroporézních gelů a bioaktivního anorganického plniva. Tento materiál bude složen z matrice, představované jednak relativně rychle degradovatelnými hydrogely na bázi methakrylátu nebo makroporézním hydrogelem na bázi přírodního kolagenu (oba typy matrice budou obohaceny kyselinou hyaluronovou). Materiál bude dále obsahovat kolagenní nebo polylaktidová nanovlákna a anorganické plnivo (přírodní nebo syntetický hydroxyapatit příp. trikalciumfosfát). Kompozity budou mechanicky testovány a bude studován vliv celých kompozitů i jednotlivých jejich složek na adhezi, růst, proliferaci, maturaci, životaschopnost a potenciální imunitní aktivaci osteogenních buněk (in vitro testy).

The objectives of the project were fulfilled, with an exception of cell testing in a dynamic cultivation system. The created high-ranking publications are dedicated to many projects, so that the quality and quantity of outputs is lower in relation to thecosts.
