Phenolic metabolism, antioxidants and stress tolerance in algae

Project goals

With respect to extensive diversity of algae, number of their secondary metabolites is high. However, they were studied mainly in economically important algae and typical phenolic metabolites of vascular plants were not extensively studied. Phenolic metabolites (flavonoids and phenolic acids) in control and stress-exposed micro-algae and activities of related proteins (phenylalanine/tyrosine ammonia-lyase and chalcon synthase) will be analyzed. Additionally, relation of phenolics to other low molecular antioxidants (ascorbate and glutathione) using specific inhibitors will be studied. Selected flavonoids will be quantified in micro- and macro-algae aimed to compare evolutionary diversification. Finally, impact of mineral nutrition on induction of phenolics and antioxidative activities of algal extracts will be assayed.

    Vzhledem k vysoké diversitě řas se v nich vyskytuje velké množství sekundárních metabolitů. Dosud však byly studovány převážně v ekonomicky užívaných řasách; typickým fenolickým metabolitům vyšších rostlin se nevěnovala rozsáhlejší studie. V rámci projektu budou sledovány fenolické metabolity (flavonoidy a fenolové kyseliny) v kontrolních a stresu vystavených tzv. mikro-řasách a bude studována aktivita souvisejících proteinu (fenylalanin/tyrosin amoniumlyázy a chalkonsyntázy. Dále budou studovány vztahy mezi fenoly a jinými nízkomolekulárními antioxidanty (askorbát a glutathion) s využitím specifických inhibitorů. Vybrané flavonoidní deriváty budou kvantifikovány v mikro- a makro-řasách s cílem porovnat evoluční diverzifikaci. Také se bude sledovat indukce fenolů vlivem minerální výživy a antioxidační aktivita extraktů řas.

    Majority of specific tasks has been met (development of a method for analysis of polyphenols in macroalgae, oxidative stress and antioxidants, polyphenols formed in the presence of Cu, Ni and Cd ions); 4 peer-reviewed papers and 1 manuscript under review have been prepared within this project. Students participated significantly in this project. No problems in financial management were identified.
