Improved growth of human skin cells on biomimetic nanofibrous matrices for active wound healing

Project goals

In this Project, the adhesion, growth and phenotypic maturation of human keratinocytes and dermal fibroblasts will be studied in vitro on novel polymeric nanofibrous membranes releasing bioactive amino acids (L-arginine), peptides, such as glycyl-L-histidyl-L-lysine (GHK), macrophage migration inhibitory factor (MIF) and antimicrobial peptides (AMPs), and also coated with cell adhesion-mediating proteins, namely fibrin, collagen and fibronectin. Bilayered constructs of keratinocytes and fibroblasts on the membranes will be created. The membranes will be made of polycaprolactone (PCL) and particularly its copolymer with polylactide (PLA-PCL) of various PLA:PCL ratios in order to regulate the mechanical properties, surface wettability and the degradation rate of the material. Also water-soluble polymers, e.g. polyvinylalcohol, polyethylene oxide and polyvinylpyrrolidon, will be explored. The potential of the meshes releasing bioactive peptides and amino acids to promote and accelerate the active wound healing will be also tested in vivo in experimental skin wound produced in rats.

    Projekt je zaměřen na studium adheze, růstu a fenotypické maturaci lidských keratinocytů a kožních fibroblastů v kulturách na nově vyvíjených polymerních nanovlákenných membránách uvolňujících bioaktivní aminokyseliny (L-arginin) a peptidy, jako je glycyl-L-histidyl-L-lysin (GHK), faktor inhibující migraci makrofágů (MIF) a antimikrobiální peptidy (AMP). Membrány budou rovněž pokryty proteiny zprostředkující adhesi buněk, jako je fibrin, kolagen a fibronektin, a poslouží k vytvoření dvouvrstevných konstruktů keratinocytů a fibroblastů na protilehlých stranách membrán. Membrány budou připraveny z polykaprolaktonu (PCL), a zejména z jeho kopolymerů s polylaktidem (PLA-PCL) s různým poměrem PLA:PCL pro regulaci mechanických vlastností, smáčivosti a degradability materiálu. Budou vyzkoušeny i polymery rozpustné ve vodě, např. polyvinylalkohol, polyetylén oxid and polyvinylpyrrolidon. Bude rovněž sledována schopnost nanovlákenných membrán uvolňujících bioaktivní peptidy a aminokyseliny podporovat a urychlovat aktivní hojení ran po experimentálně způsobeném poškození kůže u potkana.
