    Chitosan is a linear polymer, which is biodegradable, biocompatible, non-toxic and therefore predestined for use in a variety of biological, biomedical applications. However, chitosan is also a very heterogeneous material that is difficult to define. Shorter chitosan chains (COS, 2-25 units) with varying degrees of acetylation are suitable for biological applications. The usual hydrolysis of long chitosan chains by concentrated hydrochloric acid results in a heterogeneous mixture of short COS that is difficult to separate, and the kinetics of this reaction are additionally difficult to control. This project focuses on the tunable recovery of defined low molecular weight chitosan with low polydispersity by two processes: (i) deacetylation of chitin and subsequent hydrolysis of chitosan using organic/inorganic acids in solid-state assembly or (ii) alkali-catalyzed one-step mechanochemical deacetylation and depolymerization of chitin. Defined COS material will be used as multivalent, biocompatible carriers of glycomimetics and carbohydrates to target biomedically important lectins.
