Optimalizace procesu selekce a detekce kalusu a rostlin transformovaných geny dapA a phyt
Annotation in Czech
Cílem řešeného projektu je navázat na probíhající mezinárodní transformační program s aplikací phyt a dapA genů pro zvýšení nutriční hodnoty ječmene. Půjde o studium působení fyzikálních a nutričních faktorů schopných optimalizovat proces indukce somatických embryí, regenerace a selekce in vitro u vybraných genotypů. Zvládnuté metody jsou potřebné k získání a vyselektování rostlin odvozených z transformovaných buněk. Přítomnost nastřelených genů na buněčné úrovni bude ověřena histochemickou analýzou exprese GUS-aktivity. Zavedení a optimalizace molekulárních technik detekce transformovaných kalusů a rostlin phyt a dapA geny pomocí PCR analýzy a Southernovy hybridizace.
