Vizualizace tvorby kolagenu osteogenními buňkami v kulturách na vrstvách nanokrystalického diamantu
Annotation in Czech
Vrstvy nanokrystalického diamantu (NKD) s různými fyzikálně-chemickými vlastnostmi (drsnost a topografie, smáčivost a elektrická vodivost dosažená dopací bórem) budou naneseny na křemíkové a titanové substráty v mikrovlnném plasmatu metodou chemické depozice z par. Vrstvy budou použity jako substráty pro adhesi a růst kostních buněk a jejich fenotypickou maturaci, která bude studována zejména z hlediska tvorby kolagenu, hlavního proteinu extracelulární matrix kostní tkáně. Kolagen bude znázorněn a kvantifikován moderními optickými technikami, a to dvoufotonovou excitační mikroskopií a s využitím generace druhé harmonické. Produkce kolagenu a jeho vyzrávání do vláknitých struktur bude modulováno dynamickou kultivací buněk v průtokovém nebo rotačním bioreaktoru, a rovněž elektrickou stimulací buněčných kultur na vrstvách NKD dopovaných bórem. Tvorba kolagenu bude uvedena do souvislosti s dalšími ukazateli osteogenní diferenciace (aktivita alkalické fosfatázy, tvorba osteokalcinu a kostního sialoproteinu, mineralizace kostní matrix).

Project results evaluation
Formation of nanocrystalline diamond layers of different properties was studied for the use as substrates for controlled adhesion, growth and differentiation of bone cells. Collagen quantification as a marker of the maturation of cells was monitored by means of new or adapted techniques. The results were published in 16 impacted journals. Panel P108 evaluates obtained results as excellent.
