    Probádání regioselektivit u evolučně získaných epoxidhydrolas

    Cílem projektu je hlubší porozumění jedné důležité, ale dosud opomíjené vlastnosti epoxidhydrolas (EHs), a to je regioselektivita. Vztah mezi strukturou a funkcí u dvou strukturálně rozdílných EHs bude zkoumán řízenou evolucí v oblasti substrátového tunelu (tj. iterativní saturační mutagenezí a hledáním funkčních enzymů s pozměněnou regioselektivitou). Bude následovat podrobná charakterizace dvou finálních variant těchto evolučně získaných enzymů pomocí steady-state kinetiky (substrátové spektrum, enantioselektivita a regioselektivita) a počítačových metod (modelování struktury a substrate docking). Náš výzkum objasní vztah mezi strukturou vazebného místa a kinetickým rozlišením enzymového ataku při reakci vůči jednotlivým enantiomerům substrátu u EHs.Lepší porozumění regioselektivitě u EHs nám pomůže vytvořit enantiokonvergentní EHs, které jsou velice užitečnými biokatalyzátory v biotransformacích.

    During the project timeframe there were signifficant changes in the research team which negatively influenced the outcome. However, they were able to select several mutants of the enzyme which worked. The results were published in 3 solid papers. Therefore, the project can be approved as fullfilled.
