Production of ergot alkaloid during the interaction of rye plant (Secale cereale) with fungus Claviceps purpurea

Project goals

The fungus Claviceps purpurea is a naturally occurred parasite of rye. Infection of flowers initiates fungal sexual life cycle and synthesis of ergot alkaloids. Ergot alkaloids are used for preparation of various kind of therapeutics. Their biosynthetic pathway is encoded by the cluster containing 14 ORFs. The main objective of the project consists on preparation of fungus transgenic strains in order to enhance or change spectra of produced alkaloids by overexpression of selected cluster or regulation genes. Different strains of fungus and species of genus Claviceps varying in productivity will be screened for genetic variability to establish a link to ergot quality and quantity. Progression of plant infection is a crucial factor arbitrating quantity of produced alkaloids. Thus, the other aim of the project is expression profiling of genes involved in alkaloid biosynthesis and its regulation in order to map variation among different strains of C. purpurea. Subtractive hybridization will be used to reveal possible plant proteins important for susceptibility of rye to fungal pathogen.

    Produkce ergoidních alkaloidů během interakce rostliny žita (Secale cereale) s parazitickou houbou Claviceps purpurea

    Houba Claviceps purpurea je přirozeným parazitem žita, kdy infekce sporami neopýlených květu zahajuje sexuální cyklus houby během kterého dochází k produkci námelových alkaloidů. Námel slouží jako výchozí surovina k výrobě řady léčiv. Enzymy biosyntetické dráhy námelových alkaloidů jsou kódovány čtrnácti geny speciálního klastru. V rámci projektu budou připraveny transgenní kmeny nadexprimující vybrané geny klastru za účelem kvalitativní či kvantitativní změny složení námelových alkaloidů. Bude studovaná sekvenční variabilita a počty kopií jednotlivých genů u různých druhů rodu Claviceps lišících se ve spektru a kvantitě produkovaných alkaloidů. Zahájení a progrese infekce rostliny patogenem jsou klíčové pro výsledné množství produkovaných alkaloidů, proto budou sledovány změny v expresních profilech genů zapojených do biosyntézy alkaloidů a její regulace především u kmenů s různou produktivitou a transgenních kmenů. Metoda subtraktivní hybridizace bude využita pro hledání potencionálních genů v transkriptomu žita důležitých pro susceptibilitu rostlinného hostitele k patogenu.

    Aims of the project were fully met. Methods for manipulation, genetic transformation and plant interaction studies of fungus Claviceps have been introduced. Novel knowledge about the regulation and production of ergot alkaloids was obtained.
