Two-dimensional separation techniques for microscale bioanalysis

Project goals

The intention is to perform on-line coupling of two separation methods in the capillary format. Resulting two-dimensional (2D) separation is expected to have high efficiency and good detection sensitivity. High peak capacity will be achieved by a combination of electroseparation methods based on different separation principles with high degree of orthogonality. Detection sensitivity will be addressed by a laser induced fluorescence (LIF) detection in conjunction with on-line preconcentration of  the sample in the first dimension of the separation. The research will be preferably focused on temperature-gradient electrofocusing method, which is rather new method combining focusing and separation effects. Application field for 2D microscale separations isin the analysis of biosamples, such as proteomic or metabolomic samples, the inherent complexity of which is a serious challenge for established one-dimensional methods such as HPLC or capillary electrophoresis.
