    Studium genetické variability a stability dlouhodobých kultur somatických embryí smrku ztepilého

    Cílem projektu je studium genetické variability klonů somatických embryí smrku ztepilého a jejich stability během dlouhodobé kultivace in vitro pomocí molekulárních markerů. V rámci řešení projektu budou pro studium genetické variability klonůsomatickýchembryí pocházejících z různých lokalit použity metody AFLP a analýza mikrosatelitů. Výsledky budou porovnány s výsledky získanými pomocí RAPD markerů na 16ti dosud sledovaných klonech. Ke sledování genetické stability somatických embryí běhemdlouhodobé kultivace in vitro bude použito analýzy mikrosatelitů a AFLP. Dále bude otestována možnost použití GIRAFF jako metody umožňující zachycení bodových mutací. Stanovení budou prováděna jednak na průběžně odebíraných vzorcích DNA a dále navzorcích DNA z minulých 5-ti let na nichž pomocí RAPD markerů nebyla zjištěna genetická nestabilita. Vznikne tak unikátní soubor dat sledování genetické stability v průběhu osmi let.
