Role of the sex chromosome W in sex determination of Lepidoptera

Project goals

Lepidoptera possess a WZ/ZZ (felame/male) sex chromosome system or its numerical variations. However, little is known about the sex-determining mechanism in this large insect order. Our recent results, obtained by comparative genomic hybridization (CGH)in Bombyx mori, Ephestia kuehniella, and Galleria mellonella suggest an accumulation of common repetitive sequences in the W chromosome. In addition, we found a male killing activity in a W-chromosome segment of Ephestia mutants. We propose here a detailed analysis of the W chromosome in selected species by methods of molecular cytogenetics. We intend to study molecular differentiation of the W by CGH, molecular composition of the W by fluorescence in situ hybridization (FISH) mapping of probes derivedeither from Bombyx retrotransposones or from W-chromatin, and pattern of histone acetylation in W and Z.It is expected that results obtained will contribute to understanding the role of W chromosome in sex determination of Lepidoptera.

    Role pohlavního chromosomu W v determinaci pohlaví motýlů

    Lepidoptera mají WZ/ZZ (samice/samec) systém pohlavních chromosomĎ nebo jeho numerické variace. Avçak velmi málo je známo o mechanismu determinace pohlaví u tohoto velkého ýádu hmyzu. Naçe nedávné věsledky, získané komparativní genomovou hybridizací (CGH) u Bombyx mori, Ephestia kuehniella a Galleria mellonella, indikují akumulací bŘÔněch repetitivních sekvencí v chromosomu W. TéÔ jsme v segmentu W chromosomu u mutantu Ephestia prokázali pýítomnost faktoru letálního pro samčí pohlaví. Navrhujeme detailní analězu chromosomu W u vybraněch druhů metodami molekulární cytogenetiky. Hodláme studovat molekulární diferenciaci W pomocí CGH, molekulární složení W mapování sond odvozeněch z retrotransposonů B. mori či z W-chromatinu fluorescenční in situ hybridizací (FISH), a také acetylaci histonů u W a Z. Očekáváme, že získané výsledky přispějí k poznání role chromosomu W v determinaci pohlaví motýlů.
