Centre for molecular ecology of vectors and pathogens

Project goals

The main aim of the project is collaboration of top scientific institutions in parasitology and molecular biology, to increase their competitive ability and improve education of young parasitologists. Molecular interactions significant for transmission of pathogen will be studied on the models tick ? Borrelia, phlebotomus ? Leishmania and trematode ? snail. Immune interaction vector ? host will be analyzed in the context of pathogen transmission. Analysis of molecular interactions between phlebotomus midgut and surface proteins of Leishmania will elucidate the vector specificity. Genes controlling host susceptibility to the infection with Leishmania and Borrelia will be mapped. Analysis of Trichobilharzia peptidases will clarify the parasite ? host compatibility. Immune interactions snail ? trematode will be analysed. Lectin and immune interactions with Borrelia spirochetes will be studied in ticks, as well as immunomodulatorty molecules in tick saliva with the aim of development of anti-tick vaccines. Studies on molecular ecology of tick-borne pathogens will contribute to disease prevention. Vaccines based on tick concealed antigens will be developed

    Cílem projektu je podpořit spolupráci špičkových domácích parazitologických a molekulárně biologických pracovišť, zvýšit jejich světovou konkurenceschopnost a zkvalitnit výuku mladých parazitologů. Na modelech klíště - borelie, flebotomus - leishmanie, motolice - plž budou studovány molekulové interakce významné pro přenos patogena vektorem. Imunitní interakce vektora s hostitelem bude analyzována v souvislosti s přenosem patogena. Studium molekulárních interakcí mezi střevem flebotomů a povrchovými proteiny leishmanií objasní specifitu vektorů. Budou zmapovány geny kontrolující vnímavost hostitele k infekci leishmaniemi a boreliemi. Mapování peptidáz u trichobilharzií objasní kompatibilitu parazit ? hostitel. Budou analyzovány imunitní interakce trichobilharzií s mezihostitelskými plži. U klíšťat budou studovány lektinové a imunitní interakce s přenášenými boreliemi a imunomodulační molekuly klíštěcích slin s cílem vývoje vakcíny proti klíšťatům a nových léků pro léčbu alergií. Studium molekulární ekologie lymské boreliózy a klíšťové encefalitidy přispěje k jejich prevenci. Budou vyvinuty vakcíny založené na skrytých antigenech klíštěte

    Molecules of vectors (tick, phlebotomus, snail) and pathogens (Borrelia burgdorferi, tick-borne encephalitis virus, Trichobilharzia) participating in their mutual interactions were identified and characterized. The role of vector saliva in pathogen transmission was elucidated. Molecular basis of hemoglobin digestion in tick midgut was characterized. Anti-tick vaccine was patented.
