    Charakterizace a aplikace fungálních nehemových oxidas

    Cílem projektu je produkce a charakterizace nových nehemových oxidas fungálního původu pro biotechnologické aplikace. Zaměříme se zejména na tyrosinasy basidiomycet. Bude zjištěn výskyt tyrosinas u sbírkových kultur těchto hub a proveden průzkum databasígenů a proteinů s cílem najít hypotetické enzymy homologní se známými tyrosinasami, a konstruovány homologní modely nových enzymů. Vybrané enzymy budou exprimovány v heterologních hostitelských kmenech, purifikovány a charakterizovány, a bude proveden pokus krystalizovat vybraný enzym pro rentgenostrukturní analýzu. Na základě poznatků o substrátové specifitě a stabilitě budou zvoleny enzymy vhodné pro biodegradaci a detekci fenolických polutantů (hlavně chlorovaných a bromovaných fenolů, bisfenolu A,tetrabromobisfenolu A) a pro přípravu jejich metabolitů. Pro tento účel budou dále využity syntetické katalyzátory dostupné v konsorciu akce a budou porovnány produkty a účinnost těchto katalyzátorů a námi připravených enzymů.

    The project contributed to the development of methodical approaches to be used in metatranscriptomics and also delivered metagenomic and metatranscriptomic characteristics of soil biotopes with pan-european importance. Most emphasis was devoted to the analysis of sequencing data, obtained from selected ecosystems in the Czech Republic, Europe and worldwide.
