    Enzymová příprava glykosidických metabolitů biologicky aktivních látek

    Budou vypracovány krystalové struktury samotné rutinosidasy z kmene Aspergillus niger i v komplexu se substrátem. Připravíme rekombinantní kmeny Pichia pastoris nebo Escherichia coli nadprodukující pozměněnou rutinosidasu z kmene Aspergillus niger s vyšší transglykosylační aktivitou. Varianty rutinosidasy budou získány cílenou nebo saturační mu-tagenezí a řízenou evolucí. Nejaktivnější varianty budou charakterizovány na úrovni jejich primárních struktur, substrátových spekter a výše transglykosylačních a hydrolytických aktivit v různých časových intervalech reakcí s rutinem jako donorem rutinosy. Tyto rekombinantní enzymy budou použity k syntéze různých, v přírodě se nevyskytujících diglykokonjugátů na bázi rutinosy. Výsledky projektu budou publikovány v recenzovaných článcích, část výsledků by mohla být použita k podání patentové přihlášky.

    This project investigates a fungal diglycosidase - a unique rutinosidase specific for the diglycoside rutin, using a mutagenesis approach based on site-directed and saturation mutagenesis in conjunction with directed evolution. We have obtained high-resolution X-ray data and the subsequently solved first three-dimensional structure of a rutinosidase, which served for new synthetic aplications.
