    Nové aldoximdehydratasy: izolace, charakterizace a využití pro biokatalytické moduly

    Cílem projektu BIOMODUL je připravit zcela nové aldoximdehydratasy z vláknitých hub heterologní expresí příslušných genů syntetizovaných podle sekvenčních dat uložených v databázi GenBank a popsat jejich strukturu a katalytické vlastnosti. Dalším cílem je zhodnotit užitečnost těchto enzymů pro biotechnologie, konkrétně přípravu hydroxynitrilů (kyanhydrinů), aminonitrilů, ketonitrilů a dinitrilů z aldoximů, tj. ekologicky šetrnou metodou bez použití vysoce toxických chemikálií jako je HCN. Poté budou AODasy integrovány do umělých biokatalytických drah (modulů) zahrnujících nitrilasy nebo nitrilhydratasy/amidasy jako katalyzátory dalšího kroku – hydrolýzy nitrilů na příslušné karboxylové kyseliny (hydroxy-, amino-, keto- a kyanokyseliny, případně příslušné amidy (stavební bloky farmaceutických a speciálních chemikálií). Tohoto cíle má být dosaženo k termínu ukončení projektu, tj. 31.10.2017.

    The first aldoxime dehydratase from a bacterium of the genus Bradyrhizobium (soil organisms, plant symbionts) was prepared in a heterologous host. The probable role of this enzyme in the original organism is the biotransformation of natural aldoximes (defense compounds, auxin precursors). In vitro the enzyme can be used in biocatalysis for the synthesis of nitriles from aldoximes.
