Phospholipase D regulated transcriptome of systemic acquired resistence in Arabidopsis thaliana

Project goals

Project is focused on the study of phospholipid signalling in the processes leading to systemic acquired resistance, which is one of the mechanisms of plant response to pathogene attack. Basic characteristic of SAR is the early accumulation of salicylicacid (SA), both in the site of infection and in distal plant tisues. SA triggers a complex cascade of signaling events resulting in the expression of PR proteins. The aim of the project is to analyze the transcriptome in the cells of Arabidopsis thalianasuspension culture treated with SA and n-butanol (potent inhibitor of PLD) and thus reveal the group of genes dependent the activity of PLD, which is one of the key enzymes of phospholipid metabolism in plants. It seems very likely that this group willbe mostly identical with those regulated by phosphatidylinositol 4-kinase, the enzyme producing phosphatidylinositol 4,5-bisphospahate, an activator of PLD. It would implicated the close connection of both enzymes in SA pathway in Arabidopsis thaliana.

    Úloha fosfolipasy D při regulaci transkriptomu systémové získané resistence u Arabidopsis thaliana

    Projekt je zaměřen na studium úlohy rostlinného fosfolipidového signálního systému v procesech vedoucích ke spuštění systémové získané resistence (SAR), jednoho z mechanismů jímž rostliny reagují na napadení patogenem. Základním projevem SAR je akumulacekyseliny salicylové (SA) a to i v nenapadených částech rostlin vedoucí ke spuštění exprese obranných genů (PR-geny). Cílem projektu je nalézt pomocí analýzy DNA chipů v buňkách suspensní kultury Arabidopsis thaliana ošetřených SA a n-butanolem (inhibitor PLD) skupinu genů závislých na aktivitě fosfolipasy D (PLD), jednoho z klíčových enzymů metabolismu fosfolipidů u rostlin. Je pravděpodobné, že tato skupina genů bude z velké části shodná se skupinou genů závislých na aktivitě fosfatidylinositol 4 kinasy, jejíž produkt fosfatidylinositol 4,5-bisfosfát je aktivátorem PLD. To by ukazovalo na těsné spojení funkce obou enzymů v signální dráze odpovědi na SA v Arabidopsis thaliana. Dále bude možné identifikovat dosud nepopsané SA-indukované geny.
