Chromatin Regulation of Phaseolin Transcrption
Project goals
Phaseolin, a major seed storage protein of Phaseolus vulgaris is encoded by phaseolin (phas) gene. During seed development, phas is expressed at exceptionally high levels but it is fully repressed in all vegetative tissues. The establishment of a repressive chromatin structure and the presence of a nucleosome over three phased TATA boxes of the phas promoter is responsible for the lack of phas expression in vegetative tissue. Tranion from the phas promoter is enabled during early embryogenesis by a two-step process. Step 1 involves chromatin remodeling (by the B3 domain-containing tranion factor PvALF), that permits formation of a potentiated tranion complex. In step 2, tranional activation is initiated by an ABAinduced signal transduction cascade. Themain goal of this proposal is to contribute to the explanation how changes in chromatin structure over the phas promoter are orchestrated to permit tranion. Project results evaluation
Our work focused on discovering genes underlying PvALF mediated expression of the Phas promoter. Chromatin modifying enzymes, modules/clusters of genes that assist with understanding of gene-gene, gene-function interactions for chromatin remodeling during seed development were identified by next generation sequencing. This gene network is transcriptionally controlled by PvALF during embryogenesis.
