Contribution of adipocyte/macrophage fatty acid-binding protein (FABP)-4 to the pathogenesis of rheumatoid arthritis and osteoarthritis
Project goals
Annotation I: Peripheral blood and synovial fluid will be obtained from patients with rheumatoid arthritis and osteoarthritis. Synovial membrane and hyaline cartilage will be obtained from the registry of samples of the Institute of Rheumatology. Annotation II: FABP4 will be analyzed in serum, synovial fluid, synovial membrane and hyaline cartilage. Methods of immunohistochemistry, commercially available ELISA kit, Western blot and gene expression will be used for the analysis. An extracellular role ofFABP4 will be studied with stimulation experiments on cell lines of monocytes, synovial fibroblasts and chondrocytes. An impact of FABP4 on the expression of genes participating in the pathogenesis of rheumatic diseases will be studied. Annotation III: An intracellular role of increased expression of FABP4 by the cells of articular compartment will be evaluated by gene interference. A relationship of FABP4 to an inflammatory state, activity and severity of the disease will be evaluated.

Anotace I: Periferní krev a synoviální tekutina bude získána od pacientů s revmatoidní artritidou a osteoartrózou. Synoviální membrána a hyalinní chrupavka budou získány z registru vzorků Revmatologického ústavu. Monocyty budou izolovány z krve a synoviální tekutiny, synoviální fibroblasty a chondrocyty budou izolovány z příslušných tkání. Anotace II: Protein FABP4 bude analyzován v séru, synoviální tekutině, synoviální membráně a hyalinní chrupavce. K analýze budou použity metody imunohistochemické, komerčně dostupný ELISA kit, Western blot a genová exprese. Extracelulární úloha FABP4 bude studována pomocí stimulačních experimentů na liniích monocytů, synoviálních fibroblastů a chondrocytů. Studován bude vliv FABP4 na expresi genů podílejících se na patogenezi revmatických onemocnění. Anotace III: Intracelulární úloha zvýšeně exprimovaného FABP4 buňkami kloubního kompartmentu bude hodnocena pomocí genové interference. Bude hodnocen i vztah FABP4 k zánětlivému stavu, aktivitě a závažnosti onemocnění

Project results evaluation
It was shown there is increased FABP4 in rheumatoid synovial tissue, synovial fluid and blood circiulation of patient with RA. Increased amount of FABP4 and its possitive correlation with lipid profile suggest that it may be related to metabolic changes
