Changes in the profile of steroids and their neuroactive metabolites in relation to prediction of term
Project goals
The parturition is a multi-factorial process. A trigger mechanism that could be associated with the induction of parturition could be the changes of progesterone levels and, in human, particularly the changes of progesterone neuroactive metabolites. We will analyze whether the changes in the 20alpha-hydroxysteroid dehydrogenase activity could influence the term like the shifts in the balances between the neuroinhibiting 3alpha-pregnanolonone isomers and competing 3beta-isomers or sulfated antagonists. The main focus of interest will be the period from the 36th week to parturition. To our view, several metabolic steps may contribute to the catabolism of parturition-sustaining progesterone, 5beta- and 3alpha- isomers of pregnanolone in human. The obtained information will help to improve the diagnostics of endocrinopathies complicating the termination of pregnancy as the CNS disturbances. The results would enable to suggest a prediction model for the assessment of term.

Změny profilu steroidů a jejich neuroaktivních metabolitů ve vztahu k predikci termínu porodu

Těhotenství je multifaktoriální proces. Jedním z impulzů, spojených s vyvoláním porodu by mohly být změny ve hladinách progesteronu a u lidí pak zejména změny ve hladinách progesteronových neuroaktivních metabolitů. Budeme zjišťovat, zdali změny aktivity20alfa-HSD mohou ovlivnit termín porodu podobně jako posuny rovnovah mezi neuroinhibičními 3alfa-pregnanolonovými izomery (PI) a kompetujícími 3beta-izomery či sulfatovanými antagonisty. Máme zájem detailněji prozkoumat období od 36. týdne gestace do porodu. Podle našeho názoru může několik metabolických kroků přispívat ke katabolismu porod udržujících steroidů progesteronu, 5beta- a 3alfa-izomerů pregnanolonu u člověka. Uvedené informace pomohou zlepšit diagnostiku endokrinopaií komplikujících závěr těhotenství jako jsou poruchy CNS. Výsledky umožní návrh predikčního modelu pro odhad termínu porodu.
