    Vztah mezi arteriální stěnou a perivaskulární tukovou tkání v iniciální fázi aterogeneze

    Projekt je zaměřen na stanovení vztahu mezi množstvím a poměrem prozánětlivých M1 a protizánětlivých M2 makrofágů v lidské renální arterii a v přilehlé perivaskulární tukové tkáni u zdravých dárců ledvin - žen. Subpopulace makrofágů budou korelovány ke specifickým rizikovým faktorům aterosklerózy včetně reprodukčního statusu a plazmatických markerům subklinického zánětu. Přímý efekt nízkomolekulárních látek uvolňovaných tukovou tkání na efflux cholesterolu z makrofágů a adhezi a migraci monocytů bude stanoven v tkáňové kultuře.

    Project was successfully completed, all aims planned were fulfilled and usually exceeded including the publication outputs. In total 81 patients were included (planned 60), paired samples were obtained from 67 patients and in 59 samples complete analysis of all parameters could be performed. Unique results were obtained that contribute to elucidating initial phases of human atherogenesis. A simultaneous analysis of macrophages in vascular wall and corresponding perivascular fat tissue was performed and significant influence of several cardiovascular risk predictors (dyslipidemia, body fat, smoking) on macrophage polarization in human fat tissue was found. Effects of statin therapy on anti-inflammatory macrophage polarization, including verification by experimental animal study (above the project plan), were investigated.
