Construction of physical map of the hexaploid wheat D genome using the chromosome-based approach
Project goals
Dissecting the complex wheat genome to smaller parts will simplify its sequencing, analysis, and positional gene cloning. We propose to construct 1D, 4D, and 6D-specific BAC DNA library suitable for construction of physical maps. The results and materials that will be obtianed will facilitate the analysis of evolution of the hexaploid wheat genome. The new genomic resources will facilitate cloning of wheat genes underlining resistance to biotic and abiotic stresses, as well as yield and quality. The BAClibraries and physical maps will provide a basis for wheat genome sequencing.

Within the framework of COST action FA0604 and in collaboration with laboratories of Prof. B.S. Gill (KSU, Manhattan) and Prof J. Dvořák (UC, Davis) BAC libraries were prepared from wheat chromosomes 1D, 4D and 6D. The libraries were used to assemble minimum tiling path representing the chromosomes. BAC contigs were integrated with physical map of Aegilops tauschii and anchored to genetic map.
