Proteolysis-targeting chimeras (PROTACs) as novel biotechnology for cancer therapy
Original language description
The ubiquitin-proteasome system (UPS) is an important pathway in cells that mediates the metabolic recycling of proteins and their disposal. A ubiquitin is attached to the e-amino group of a lysine amino acid residue on the amino-terminus in a substrate protein and starts polyubiquitination of this protein. This process involves three enzymes (E1 ubiquitin-activating enzyme, E2 ubiquitin-conjugating enzyme, and E3 ubiquitin ligase). The C-terminus of each ubiquitin is linked to a specific lysine residue (Lys48) of the previous ubiquitin if the substrate protein is destined for degradation in proteasome.
