    Detection of the mechanism of the resistance to atrazine in a Czech Chenopodium album biotype

    A Czech biotype of common lambsquarter (Chenopodium album L.) resistant to atrazine was used in this study. The resistance to atrazine was investigated using chlorophyll fluorescence and photochemical activity of isolated chloroplasts (Hill reaction) assays. The molecular basis of resistance was determined by sequence analysis of the psbA gene. A region of the psbA gene encoding D1 protein of Photosystem 2 (PS2) was amplified, sequenced and compared between the resistant and susceptible biotypes. A mutation in the psbA gene that coded for an amino acid substitution of glycine for serine at residue 264 of the D1 protein was identified. This mutation is the most likely the cause for atrazine resistance in the Czech biotype of lambsquarter.

    V této studii byl použit český biotyp merlíku bílého (Chenopodium album L.) rezistentní vůči atrazinu. Rezistence byla zkoumána pomocí fluorescence chlorofylu a fotochemické aktivity izolovaných chloroplastů (Hillova reakce). Molekulární podstata rezistence byla stanovena na základě analýzy sekvence genu psbA. Část genu psbA kódující D1 protein fotosystému 2 byla amplifikována, sekvenována a porovnána mezi rezistentním a citlivým biotypem. Byla identifikována mutace v genu psbA kódující substituci aminokyseliny glycin za serin v pozici 264 proteinu D1. Tato mutace je s největší pravděpodobností příčinou rezistence českého biotypu merlíku bílého vůči atrazinu.
