Embryogenic responsibility of selected genotypes of Brassica carinata A. Braun to microspore culture

  • Original language description

Microspore culture has been used to produce desirable meiotic recombinants in numerous Brassica species. However, the utilization of these recombinants relies on inefficient genome doubling procedures to generate fertile doubled haploids plants. Therefore, it represents a simple and rapid procedure to generate fertile doubled haploids in Brassica carinata. The effects of colchicine, trifluralin and oryzalin on microtubule depolymerization and chromosome doubling in embryogenic microspore cultures of Brassica carinata genotypes were examined. The direct application of these microtubule inhibitors to microspore cultures resulted in the recovery of fertile doubled haploids plants. The application of 10?M trifluralin during 24hrs of microspore culture wasfound to be the superior method for doubled haploid production. Similarly, all tested concentrations of colchicine and oryzalin significantly increased doubled haploid production in comparison with control variants.

Embryogenní responzibilita vybraných genotypů Brassica carinata A. Braun v mikrosporové kultuře
