    The relationship between accmulation of cold regulated proteins and frost tolerance in wheat

    The objective of this study was to use reciprocal near-isogenic lines (NILs) for the Vrn-A1 locus from spring habit cv. Manitou (Vrn-A1) and winter habit cv. Norstar (vrn-A1) of wheat (Triticum aestivum L.) to determine the influence of the Vrn-A1 locuson the expression patterns of dehydrins at the translation level using protein gel blot analysis. Anti-dehydrin and anti-WCSP1 antibodies used for Western blotting allowed the detection of the wheat dehydrins and WCSP1 in crowns and leaves. The genotypeswere cold-acclimated at 6 °C for 0, 2, 14, 21, 35, 42. 56 and 70 days. The level of WCS120 and WCSP1 correlated with the level of frost tolerance of the genotypes. Higher concentration of dehydrins and WCSP1 were found in the crowns compared to the leaves. Moreover, our results proved strong influence of VRN-A1 locus on frost-tolerance and the accumulation of cold regulated proteins.

    Cílem této studie bylo užití reciprokých blízce isogenních linií (near-isogenic lines) v lokusu Vrn-A1 odvozených od jarního kultivaru pšenice Manitou (Vrn-A1) a ozimého kultivaru Norstar (vrn-A1) za účelem stanovení vlivu lokusu Vrn-A1 na expresi dehydrinů na translační úrovni (imunobloty). Protilátky proti dehydrinům a WCSP1 umožnily detekci pšeničných dehydrinů a WCSP1 v odnožovacích uzlech a v listech. Genotypy byly otužovány při teplotě 6 °C po dobu 0, 2, 14, 21, 35, 42, 56 a 70 dnů. Úroveň akumulace WCS120 a WCSP1 korelovala s dosaženou úrovní mrazuvzdornosti. Vyšší koncentrace dehydrinů a WCSP1 byly nalezeny v odnožovacích uzlech ve srovnání s listy. Navíc naše výsledky ukázaly silný vliv lokusu VRN-A1 na mrazuvzdornost a na akumulaci COR proteinů.
