To evaluate most reliable method for European stone fruit yellows (ESFY) phytoplasma detection, apricot and peach trees infected with ESFY were evaluated for symptom expression. Biological indexing was carried out on the peach indicator GF-305 and molecular detection by PCR was carried out with different primer pairs. Some false positive reactions in nested PCR assays were obtained, but also the biological indexing was not reliable because of many negative results from symptomatic trees. The presence ofspecific and typical symptoms on leaves is still the most reliable criterion for detection of ESFY in apricot and peach trees grown in the Czech Republic.
