Automated integrative high-throughput phenotyping of plant shoots: a case study of the cold-tolerance of pea (Pisum sativum L.)
Original language description
In this the high-throughput method for automated screening of cold tolerant pea (Pisum sativum L.) cultivars was designed. Two contrast cultivars were screened simultaneously in an automated way with throughput of 16 plants per hour for i) growth of theaerial parts by RGB imaging and ii) for the efficiency of photosynthesis by chlorophyll fluorescence imaging. We demonstrated that the presented integrative approach can provide deeper insight into the physiological base of cold-acclimation. Data from both analytical tools pointed to significant differences in the growth and photosynthesis of tested cultivars, and indicated that cultivars use different strategies for cold acclimation differing in number of open PSII reaction centers, their maximal photosynthetic quantum yield in light and quantum yield of constitutive non-light induced dissipation processes. The reliability of the screening was verified by independent measuring of the fresh weight of the shoots and by Chl fluorescence m
