Original language description
Populus x canescence (grey poplar) belongs among fast-growing woody plant species. For its unique phenotype traits observed in floodplain forest in the South Moravia region of the Czech Republic and a relatively rapid growth in in vitro as well, approximately 4-6 weeks of micropropagated plants were used to test the most effective culture conditions for a long-term conservation under ultra-low temperature. Firstly, excised apical and nodal segments were followed in their growth to determine suitable plant material for cryopreservation. Apical segments exhibited more steady developmental progress opposite to unbalanced growth of nodal segments therefore modified vitrification protocol (80% PVS3 cryoprotectant) was applied into the apical segments of 2 mm length.
