Identification of suitable reference gene for quantitative transcription analysis (RT-qPCR) of Fusarium culmorum genes in infected barley plants
Original language description
Based on the RTqPCR aynalysis of reference genes transcription in infected barley in greenhouse and field conditions, and during the malting, we have confirmed that ubiquitin (UBC) is a suitable reference gene for normalization transcription of Fusarium culmorum genes in barley infected with the pathogen. We found a positive correlation between the transcription of the three genes and the growth of F. culmorum DNA in the affected tissues. Since the selection of a suitable reference gene is essential for measuring transcription and is to be verified not only for different parts of plants and related species, we also recommend testing whether this RG has the ability to reflect changes in the pathogen content of the infected plant tissue.
