Original language description
Garlic is a valuable source of health-promoting compounds, including secondary metabolites rich in sulfur such as cysteine sulfoxides (CSO). In this study, a new qRT-PCR assay is presented, analyzing the transcription of two genes encoding key enzymes of CSO biosynthetic pathway (cysteine synthase and alliinase) in developing garlic. In addition, a set of three reference genes (RGs) is presented which are suitable for normalization of the transcription of the described genes of interest. Using newly introduced qRT-PCR assays unique to cysteine synthase (chloroplast and cytosol specific), alliinase, and using RGs, we have shown that the corresponding genes have been actively transcribed throughout the whole life cycle with a peak in the germination phase. Since the content of sulfur-rich metabolites in garlic bulbs is based, among other factors, on the transcription of genes involved in the sulfur metabolism pathways in the leaves, this test can be used to examine the impact of agricultural practices or the environment on the content of sulfur metabolites in harvested garlic.
