The aim of the National Cryobank at the Czech Agrifood Research Center is to serve as a safe backup facility for the preservation of vegetatively propagated plants. One of its primary objectives is to preserve healthy plant; therefore, establishing in vitro stock plant material is a crucial step. However, introducing woody crops into in vitro conditions presents challenges due to potential contamination from surface-associated pathogens. This study aimed to evaluate the feasibility of initiating in vitro cultures of selected fruit tree species using two different sources: sprouting shoots collected from an orchard and dormant buds harvested during the winter period. Surface sterilization was performed using chlorine-based disinfectants. Subsequently, the growth and multiplication of the cultures in bioreactors utilizing a temporary immersion system (TIS) were monitored. Experiments were conducted with specific varieties of stone fruits and berry fruits. The study assessed the development of in vitro cultures and the response of different fruit tree species within the bioreactor system. The proposed approach highlights the potential of integrating TIS into the in vitro cultivation of selected fruit tree species.
