The aim of this contribution is to determine the influence of different agricultural practices on carbon sequestration. Therefore, for the purposes of this contribution we chose two locations (out of total of six in the project) with the widest possible spectrum of agricultural treatments. First field was close to Brno (chernosol area), where these types of practices were compared: regenerative agriculture, conventional ag., regenerative ag. with addition of compost, regenerative ag. with mineral fertilization and regenerative ag. with addition of digestate. The second field was close to Častrov (highland in the centre of Czech Republic with cambisol soil typel), where conventional ag. treatment, addition of compost and perennial grassland were compared. Soil samples were then taken from a depth of 0-5 cm and 20-25 cm. To confirm or refute the assumption that the organic carbon content and quality may differ in soil aggregates of different sizes, four fractions of soil aggregates (0-1 mm, 1-2 mm, 2-5 mm and larger than 5 mm) were separated from air-dried soil samples. The stability of the aggregates was then determined on these fractions using the WSA (Water Stable Aggregates) index. At the same samples, organic carbon and DRIFT spectra (Diffuse Reflectance Infrared Fourier Transform Spectroscopy) were determined. DRIFT spectra were used to express the quality of soil organic matter based on the ratio of aliphatic and aromatic functional groups. Preliminary results show that the content of soil organic matter and the stability of aggregates are significantly influenced by different agriculture treatments. Increased organic carbon content and greater stability of aggregates were found when plowing plant residues or using organic fertilizers. Furthermore, it is evident that the stability of aggregates increases with increasing aggregate size. The effect of aggregate size on the quantity and quality of organic matter was also noted in some variants.
