Original language description
Influence of growth regulators on somatic embryo initiation and histological analysis were determined in Rhododendron catawbiense Michx. 'Grandiflorum' and Dendranthema grandiflorum 'Reagan'Somatic embryos in rhododendron were initiated in the presence of cytokinins TDZ (thidiazuron) 5 mg.l-1 and adeninsulphate 40 mg.l-1 after 6 weeks of culture. In cv. Reagan somatic embryos were induced after transfer from medium supplemented by 2,4-D (2,4-dichlorophenoxyacetic acid) 1 mg.l-1 and BAP (6-benzylaminopurine) 0.2 mg.l-1 on medium without hormones. Culture full-strenght MS medium (Murashige and Skoog, 1962) was used in both cultivars. Tissues for histological studies were vacuum-infiltrated with FAA, dehydrated in a graded series of ethanol and embedded in paraffin wax. The embryogenic mass and somatic embryos were mostly derived from several morphologically competent cells
