Different evolutionary trails in the related genomes Cricetus cricetus and Peromyscus eremicus (Rodentia, Cricetidae) uncovered by orthologous satellite DNA repositioning

  • Original language description

Centromeric repetitive sequences from chromosome 4 (CCR4/10sat) of Cricetus cricetus (CCR, Cricetidae, Rodentia) were isolated using the laser microdissection, followed by DOP-PCR amplification and labelling. The probe was mapped by fluorescent in situ hybridisation onto C. cricetus and another member of Cricetidae, Peromyscus eremicus. The centromeric sequences showed to be present in another C. cricetus chromosome (CCR10) besides CCR4. Moreover, these almost chromosome-specific sequences revealed to be present in the P. eremicus genome, and most interestingly, displaying a ubiquitous scattered distribution throughout this karyotype. In both species, a co-localisation of CCR4/10sat with constitutive heterochromatin was found. The presence of these orthologous sequences in both genomes is suggestive of a phylogenetic proximity.

Různé cesty evoluce u příbuzných druhů Cricetus cricetus and Peromyscus eremicus (Rodentia, Cricetidae) stanovené přesunem orthologní satelitní DNA

  • Czech description

Centromerické repetitivní sekvence z chromozomu 4 (CCR4/10sat) Cricetus cricetus (CCR, Cricetidae, Rodentia) byly izolovány laserovou mikrodisekcí, amplifikovány pomocí DOP-PCR a značeny. Sonda byla mapována fluorescenční in situ hybridizací na chromozomy C. cricetus a dalšího zástupce čeledi Cricetidae, Peromyscus eremicus. Centromerické sekvence byly kromě CCR4 přítomny i na dalším chromozomu (CCR10) C. cricetus. U P. eremicus sonda vytvářela rozptýlený signálna všech chromozomech. U obou druhů byla nalezena společná lokalizace repetitivní CCR4/10sat s konstitutivním heterochromatinem. Přítomnost této orthologní sekvence u obou genomů naznačuje jejich fylogentickou příbuznost.
