Changes in functional parameters of cryopreserved boar spermatozoa capacitated in vitro

  • Original language description

The aim of this study was to assess functional status of spermatozoa after cryopreservation, separation and capacitation to characterize the influence of these procedures on spermatozoa functional parameters. Boar semen were frozen-thawed, motile spermatozoa were separated on Percoll-gradient and capacitated by 1mM caffeine. Motility, viability and chromatine integrity of spermatozoa were evaluated by phase contrast, Annexin V-FICT apoptosis kit and SCSA assay respectively. The high cryotolerant- (HCT)and the low cryotolerant- (LCT) boars were classified in previous experiments. There were no differences in motility and viability spermatozoa between both boar groups before freezing but after thawing, the significantly higher motility and viability were found for HCT- compared with those for LCT-boars. Separation was more effective in the LCT- than in HCT- boars.. In both groups, DNA of spermatozoa was highly stable during cryopreservation, separation and capacitation.
