    58TH CONFERENCE ADVANCES IN ORGANIC, BIOORGANICAND PHARMACEUTICAL CHEMISTRY "LIBLICE 2024" SPINDLERUV MLYN - HARMONY CLUB HOTEL, 7 - 9 NOVEMBER 2024 – poster. In our work we introduce new series of half-sandwich Ru(II) and Os(II) complexes with the general formula [M(η6 pcym)Cl(L)]PF6, involving ethane-1,2-diamine-based Schiff bases bearing two terminal polycyclic aromatic hydrocarbon (PAH) substituents (Fig. 1). These complexes were fully characterized (e.g. multinuclear NMR, mass spectrometry or HPLC), and their physicochemical properties, including solution stability and DNA interactions, were examined. The results showed that these complexes are stable in the presence of water under the mimicked physiological conditions. The primary focus of our work was on their biological activities, with potential applications as novel cytostatic agents. The complexes exhibited stronger cytotoxic effects against various lung cancer cell lines, including those resistant to cisplatin (CDDP), when compared to CDDP. Unlike CDDP, these complexes did not cause alterations in the cell cycle but were found to impact mitochondrial function, contributing to cell death Analysis of the expression of stress-related genes revealed the ability of used complexes to induce endoplasmic reticulum stress and oxidative stress. In all cell-based in vitro experiments, Os(II) complexes were more potent than Ru(II) ones. It can be concluded that the studied Ru(II) and Os(II) complexes with PAH substituents represent a promising group of compounds with significant anti-cancer potential and with different mode of action than CDDP.
