    Model Of Telomere Shortening And Immunosenescence

    The immune system is the defence mechanism aimed to combat pathogens as well as damaged self-cells. Age-associated immune dysfunction also termed 'immunosenescence', is a common phenomenon responsible for susceptibility to numerous diseases (Hornsby, 2007; Ponnappan & Ponnappan, 2011). Intensive research has been conducted recently to understand the phenotypic and functional defects caused by aging within the human immune system. However, a clear understanding of the immune system aging and its molecular and cellular mechanisms remains mostly unknown. One of the major hallmarks of aging process is telomere shortening. Functional component of human telomerase reverse transcriptase (hTERT) was silenced in the cell line of induced pluripotent stem cells using siRNA. Changes in hTERT expression were analyzed using qPCR and changes in total length of telomeres were analyzed from genomic DNA. We have established model where we can experimentally modulate the control of telomere elongation mechanism in human induced pluripotent stem cells. This tool can be used for further analysis of molecular mechanism of aging processes.
