    Standardized hiPSCs culture – vitronectin coating for cGMP compliant protocol

    Background. Human induced pluripotent stem cells (hiPSCs) hold great promise for disease modeling, drug screening, toxicology testing, regenerative medicine and more. An important task for stem cell research translation into the clinic is culturing cells in completely defined conditions. Currently, many researchers culture hiPSCs on a surface coated with Matrigel(R), poorly defined protein mixture of animal origin. Alternatively, vitronectin can be used. Methods. We cultured two hiPSCs lines in mTeSRTM1 cell culture medium on a vitronectin-coated surface and compared their pluripotent markers expression and differentiation capacity in vitro to cells cultured on Matrigel(R) coated surface. Results. hiPSCs cultured on both surfaces expressed pluripotent markers and differentiated into all three germ layers. Conclusion. In this study, we tested culture method in completely defined conditions, thus compatible with current good manufacturing practice (cGMP). We demonstrate that vitronectin is a suitable Matrigel(R) replacement
