This correspondence highlights the role of automation in physiologically based pharmacokinetic (PBPK) modeling, particularly for complex biologics models. It references the approaches of Sepp et al. (2019) and Liu et al. (2024), who implemented automated code generation using MATLAB and the Ubiquity R-package, respectively, to manage multi-compartment tissue models including vascular, endothelial, endosomal, and interstitial sub-compartments. Automation improves model accessibility, reproducibility, and error reduction, especially for large-scale models, while manual scripting remains valuable for smaller models, teaching, and understanding model mechanics.
