cDNA coding for CBP was cloned under control of metallothionein promoter of the pMT-IRES-CD4 expression vector. The internal ribosomal entry site (IRES) unit of the vector allows translation of the CD4 marker gene from a dicistronic mRNA. Following transfection of BM2 cells with the pMT-CBP-IRES-CD4 plasmid the stable transfectants inducibly expressing CBP were selected and cloned by limiting dilution. We tested inducible CBP expression in BM2CBP cells and studied the effects of CBP on the phenotype ofthe BM2 cells.
