In the present study, a series of 119 human cancers were immunostained using a highly sensitive technique that detects the low levels of wild-type protein expressed in normal cells. TP53 gene status was also assessed using FASAY and in selected cases bydirect sequencing. The transcriptional activity of TP53 was determined by staining for two downstream target genes, p21(WAF1) and MDM2, and statistical correlations between MDM2/p21(WAF1) and p53 were found in tumours with wild-type, but not mutant TP53.Measurement of staining for p53 and MDM2 accurately identifies the TP53 status of tumours.

Rozlišení funkčního a nefunkčního p53 u lidských nádorů s využitím immunohistochemické detekce p53 a MDM2

V prezentované studii jsme provedli imunohistochemickou analýzu 119 lidských nádorů s využitím velmi senzitivní metody, která umožnila detekovat velmi nízké hladiny nemutované formy p53 u normálních buněk. Status genu TP53 byl rovněž analyzován pomocí metody FASAY a ve vybraných případech robvněž přímým sekvenováním genu TP53. Transkripční aktivita p53 byla stanovena imunohistochemickou detekcí dvou cílových genů p21(WAF1) a MDM2. Byly nalezeny statistické korelace mezi expresí MDM2/p21(WAF1) a p53 pouze u nádorů s nemutovanou formou p53.Analýza barvení p53 a MDM2 přesně identifikuje status TP53 u nádorů.
