    Cardioprotective effects of a novel iron chelator, pyridoxal 2-chlorobenzoyl hydrazone, in the rabbit model of daunorubicin-induced cardiotoxicity

    In this study the cardioprotective effects of the novel lipophilic iron chelator against chronic anthracycline cardiotoxicity were investigated. Using 10 mg/kg of the chelator administered 30 min before the every daunorubicin dose the significant cardioprotection was obtained (as shown by prevention of cardiac-related deaths, improved functional, biochemical and morphological parameters). On the other hand escalation of the dose to the 25 mg/kg did not improved cardiovascular parameters, on the contrary, the cardioprotective potential was lost and early extracardiac toxicity was observed.

    V této studii byly sledovány kardioprotektivní účinky nového chelátoru železa vůči antracycklinové kardiotoxicitě. Chelátor podaný v dávce 10 mg/kg 30 min před daunorubicinem navodil signifikantní kardioprotekci (jak je patrno z prevence kardiální mortality, zlepšení funkčních biochemických a morfologických parametrů). Na druhou stranu, zvýšení dávky na 25 mg/kg nevedlo ke zlepšení kardiovaskulárních parametrů, naopak, kardioprotektivní potenciál vymizel a byla pozorována časná extrakardiální toxicita.
