    Yohimbine stability: analysis of its several decades-old pharmaceutical products and forced degradation study

    Yohimbine is an indole alkaloid traditionally used as an aphrodisiac and currently present in various pharmaceutical preparations and herbal dietary supplements. Despite the long history of yohimbine use, stability data for yohimbine in pharmaceutical products remain limited. In this study, we investigated the long-term stability of yohimbine by analyzing five several decades-old pharmaceutical products (three solid formulations and two injectable solutions) dating from the 1930s to the 1970s. Reversed-phase HPLC with UV and tandem mass spectrometric detection was used for compound identification and quantification. Additionally, a forced degradation study was conducted to evaluate the hydrolytic and oxidative stability of yohimbine under various conditions. The results confirm the remarkable stability of yohimbine in solid formulations. In contrast, significant degradation leading to the formation of yohimbinic acid was observed in acidic injectable solutions.
