Original language description
The authors described preparation and construction of a novel form of nanofiber scaffold which is loaded with immunosuppressive drug cyclosporin A (CsA). The incorporation of CsA did not influence the diameter, shape, porosity or uniformity of the nanofibers. This scaffold has an optimal kinetic of drug release and can serve for a local immunosuppression of immune reactions. The covering of skin allografts with CsA-loaded nanofibers significantly attenuated the expression of the genes for the proinflammatory cytokines. The CsA-containing nanofibers can be seeded with stem cells and thus can be used for cell transfer in regenerative medicine. The results suggest that CsA-loaded electrospun nanofibers prepared from the biodegradable polymer poly(L-lacticacid) can serve as effective drug carriers for the local/topical suppression of an inflammatory reaction and simultaneously as scaffolds for cell transfer for tissue repair and regeneration.
