argetingofendomembranetransportcontainersisoftheutmostimportanceforproperlandplantgrowthanddevelopment.Giventheimmobilityofplantcells,localizedmembranevesiclesecretionandrecyclingareamongstthemainprocessesguidingpropercell,tissueandwholeplantmorphogenesis.Cellwallbiogenesisandmodificationaredependentonvectorialmembranetraffic,notonlyduringnormaldevelopment,butalsoinstressresponsesandinplantdefenseagainstpathogensand/orsymbiosis.Itissurprisinghowlittleweknowabouttheseprocessesinplants,fromsmallGTPaseregulationtothetetheringcomplexesthatactastheireffectors.Tetheringfactorsaresingleproteinsorproteincomplexesmediatingfirstcontactbetweenthetargetmembraneandarrivingmembranevesicles.Inthisreviewwefocusonthetetheringcomplexesofthebest-studiedplantmodel-Arabidopsisthaliana.Genome-basedpredictionsindicatethepresenceofallmajortetheringcomplexesinplantsthatareknownfromahypotheticallasteukaryoticcommonancestor(LECA).TheevolutionarymultiplicationofparalogsofplanttetheringcomplexsubunitshasproducedthemassivelyexpandedEXO70family,indicatingasubfunctionalizationoftheterminalexocytosismachineryinlandplants.Interpretationoflossoffunction(LOF)mutantphenotypeshastoconsiderthatrelated,yetclearlyfunctionally-specificcomplexesoftensharesomecommoncoresubunits.Itisthereforeimpossibletoconcludewithclaritywhichversionofthecomplexisresponsibleforthephenotypicdeviationsobserved.Experimentalinterestintheanalysisofplanttetheringcomplexesisgrowingandwehopetocontributewiththisreviewbyattractingevenmoreattentiontothisfascinatingfieldofplantcellbiology.
