    Association of interleukin-8 gene variability with recurrent aphthous stomatitis

    Objectives: Variability in the interleukin-8 gene (IL-8), encoding a proinflammatory chemokine, was recently associated with Behçet's syndrome, disease with recurrent oral ulcers as one of its symptoms. The aim of this study was to analyse four single nucleotide polymorphisms (SNPs) in IL-8 and its receptor 2 (CXCR2) in patients with recurrent aphthous stomatitis (RAS) and healthy controls. Methods: Totally, 238 subjects were enrolled in this case-control study: 64 patients with RAS and 174 healthy controls were genotyped for SNPs in IL-8 (-251A/T, +396T/G, +781C/T) and CXCR2 (+1208C/T) by a method based on PCR using 5´ nuclease TaqMan assays. Results: The allele and genotype frequencies of studied polymorphisms between RAS patients and healthy controls were similar, with the exception of statistically significant differences in IL-8 +781 TT vs. CC+CT genotype frequencies between both groups. However, IL-8 A(-251)/G(+396)/C(+781) haplotype was significantly more frequent in patients with RAS (P&lt;0.02; OR=4.2; 95% CI=1.3-13.8) than in healthy controls. Conclusions: Our study indicates that the IL-8 gene variability may affect susceptibility to RAS in the Czech population.
