HPLC (high performance liquid chromatography) in the HILIC mode (hydrophilic interaction chromatography) is currently very often used in pharmacy for the separation and determination of polar analytes on polar stationary phases (in our case saccharides on the polyol stationary phase). However, since carbohydrates do not contain a suitable chromophore in their structure, a conventional UV detector is unusable for the determination, a refractometric detector would require isocratic elution, and therefore an evaporative light scattering detector (ELSD) can be used to advantage. An HPLC-ELSD method was developed for milk sugar profiling. Lactose was determined and quantified in selected commercially available infant formula samples.
