DNA binding studies of antitumor effective trans-platinum complexes with planar ligands have shown that they form much higher amount of interstrand cross-links than cisplatin. Interstrand cross-linking are formally equivalent to that of cisplatin but different from their direct ineffective analog, transplatin. This result shows that a simple chemical modification of the structure of inactive compound alters its DNA binding site into an addduct of active drug.
